ABSTRACT

OBJECTIVE To study the vasorelaxant effects and mechanism of polyphenol compound 2,4′,5′-trihydroxy-5,2′- dibromo-diphenyl-methanone(LM49)on isolated aortic rings of rats. METHODS Thoracic aortic vascular rings of rats were collected. Using the diastolic rate as index , the effects of different concentrations of LM 49 on endothelium-intact and endothelium-denuded aortic rings pre-contracted by norepinephrine (NE,1×10-6 mol/L)or KCl (60 mmol/L)were investigated. After pre-culturing vascular rings by nitric oxide synthase inhibitor L-nitro-arginine methyl ester (L-NAME,0.1 mmol/L) and cyclooxygenase inhibitor indomethacin (1×10-5 mol/L),as well as pre-culturing vascular rings by 4 potassium channel blockers [BaCl 2(0.1 mmol/L),tetraethylammonium(TEA,5 mmol/L),4-aminopyridine(4-AP,0.1 mmol/L)and glibenclamide (1×10-5 mol/L)],the vasorelaxant effect of different concentrations of LM 49 on the vascular rings were investigated by using the same method. With the percentage of vasoconstriction as the index ,using KCl (60 mmol/L),NE(1×10-6 mol/L),calcium channel blocker verapamil (1×10-6 mol/L)and sarcoplasmic Δ 基金项目 重大新药创制国家科技重大专项 (No.2018ZX097- reticulum Ca 2 +-adenosine triphosphate (ATP) enzyme pump inhibitor thacarotene (TG,1×10-6 mol/L)to induce the release of calcium in vascular rings in the absence of calcium. CaCl was added cumulatively ,and the effect of LM 49 on the cxyw06,vasoconstriction caused by calcium influx induced by CaCl 2 was investigated. RESULTS 3×10-6,5×10-6,1×10-5 mol/L LM49 had a significant relaxation effect on NE and KCl precontracted vascular rings (P<0.01); whether the endothelium was removed or not had no significant effect on the vasodilation of LM 49(P>0.05). After L-NAME ,indomethacin, TEA and 4-AP was pre-incubated ,different concentrations of LM 49 had no significant effects on aortic rings precontracted by NE (P>0.05). Glibenclamide and BaCl 2 could inhibit the vasorelaxant effects of LM 49 on aortic rings precontracted by NE (P<0.01). In the absence of calcium ,LM49 could inhibit the contraction caused by calcium influx induced by accumulated CaCl 2 after pre-incubation with KCl ,NE,verapamil and TG (P<0.01). CONCLUSIONS LM49 evokes significant relaxation of isolated aortic vascular rings without endothelium dependence ;the mechanism of which is inducing ATP-sensitive potassium channel , inward rectifier potassium channel open and restraining extracellular Ca 2 + influx via voltage-gated calcium channel , receptor-operated calcium channel and store-operated calcium channel.

ABSTRACT

We detected the isoniazid resistance in clinical Mycobacterium tuberculosis isolates by high-resolution melting (HRM) curve analysis and assessed the application value of the assay. The isoniazid resistance of 49 M.tuberculosis isolates preserved in laboratory was analyzed by the drug sensitivity test (traditional proportion method).Further analysis was made on the sequencing of the isoniazid resistance determining region in these test strains, and their mutation sites were screened.Specific primers used in the HRM curve analysis were designed based on the screened mutation sites,DNA mutations were assayed in the isoniazid-resistant gene determining region by the HRM curve analysis, and an assessment was made of the detection efficiency of the assay in isoniazid resistance in M.tuberculosis.Results of the drug sensitivity test (proportion method) showed that,of the 49 test strains, there were 20 isoniazid-resistant strains,29 isoniazid-sensitive strains. Results of the sequencing analysis showed that:1) KatG gene had four mutation patterns,i.e.,point mutations at site 234,at sites 234 and 315,at sites 234 and 463,and at sites 234,315 and 463;2) there were three mutations were detected in inhA gene,i.e.,mutations in inhA-8,-15 and-152.Analysis of gene mutation in drug-resistant strains found that of the 20 isoniazid-resistant strains,11 (55 %) were mutated at codon 315 of KatG gene;6 (30%) were mutated in inhA-15 (4/20),-8 (1/20) and-153 (1/20) of inhA gene;two (10%) were mutated at codon 315 of KatG gene and in inhA-15;in one strain (5%),no mutation was detected in KatG and inhA genes.Through the gene mutation detection, the sensitivity and specificity of isoniazid resistance in M.tuberculosis were 95 % and 100 %,respectively. Results of HRM curve analysis of drug-resistance gene mutations in test strains showed gene mutations were present in 18 strains and absent in 24 ones;referring to DNA sequencing results, the sensitivity and specificity of the assay were 94.7% and 80%,respectively.Judged by mutations as drug-resistance via the HRM curve analysis,19 resistant and 24 sensitive strains were tested. With the drug sensitivity test results by the proportion method as controls, the sensitivity and specificity of the assay were 95 % and 82.76 %,respectively.Use of the HRM curve in the detection of resistance of M.tuberculosis to isoniazid is characterized by good sensitivity and short time consuming, and has certain value in the rapid diagnosis of isoniazid-resistant tuberculosis.

ABSTRACT

AIM: To observe the expression of corticotropin releasing hormone (CRH) within the paraventricular nucleus of hypothalamus (PVN) and to explore the relationship between the activated CRH-containing neurons and sympathetic activity in rats with heart failure (HF).METHODS: Healthy male Sprague-Dawley (SD) rats were subjected to coronary artery ligation to induce HF, and chronic intracerebroventricular (ICV) infusion was performed by osmotic pump for 4 weeks. The rats in sham group and HF group were given vehicle (VEH;artificial cerebrospinal fluid 0.25 μL/h).The rats in HF plus treatment group were treated with CRH competitive inhibitor αh-CRH (15 mg/h).Meanwhile, the Lewis rats and Fischer 344 rats for control study also underwent coronary ligation to induce HF or sham surgery. After 4 weeks, left ventricular end-diastolic pressure (LVEDP) and maximum positive/negative change in pressure over time (±dp/dtmax) were determined. The right ventricular-to-body weight (RV/BW) and lung-to-body weight (lung/BW) ratios were calculated. The renal sympathetic nerve activity (RSNA) was recorded and the plasma norepinephrine (NE) level was measured. The expression of CRH in the PVN combined with the plasma adrenocorticotrophic hormone (ACTH) levels were measured.RESULTS: Compared with the sham-SD rats, the HF-SD rats had a greater number of CRH positive neurons in the PVN (accordingly the plasma ACTH levels were increased), accompanied by decreased ±dp/dtmax and increased RSNA, plasma NE, LVEDP, lung/BW and RV/BW.However, ICV treatment with αh-CRH attenuated these changes in the HF-SD rats (P<0.05).Compared with the sham-Fisher 344 rats, the HF-Fisher 344 rats also had a greater number of CRH positive neurons in the PVN (accordingly the plasma ACTH levels were increased). In addition, they had significantly increased RSNA and plasma NE level, higher LVEDP, RV/BW and lung/BW, and lower ±dp/dtmax (P<0.05).Compared with the SHAM-Lewis rats, the HF-Lewis rats had not significantly changed in the above parameters.CONCLUSION: In CHF, the CRH-containing neurons in PVN are activated, thus aggravating cardiac function by increasing sympathoexcitation.

ABSTRACT

Background:Functional gastrointestinal disorders(FGIDs)is a common disease of digestive system which severely affects the quality of life. Recently, more and more attentions have been paid to the relationship between FGIDs and psychological factors. Aims:To investigate the relationship between FGIDs and psychological factors and related treatment. Methods:A total of 270 patients with FGIDs admitted from Sept. 2013 to Sept. 2014 at Anhui No. 2 Province People’s Hospital were enrolled. Hamilton Anxiety Scale(HAMA)and Hamilton Depression Scale(HAMD)were used to evaluate the mental state of patients, and symptom severity of digestive system was scored. Patients with psychological abnormalities received conventional therapy to relieve digestive symptoms, and those who failed from conventional therapy were assigned into refractory group and others were assigned into response group. Patients in refractory group were further divided into conventional therapy group and combined therapy group for receiving paroxetine in addition to conventional therapy. Results:The rates of anxiety and depression were 61. 5%(166 / 270)and 57. 0%(154 / 270),respectively. The scores of HAMA and HAMD were positively correlated with the score of digestive symptoms(r = 0. 585,P < 0. 01;r = 0. 584,P <0. 01)in patients with anxiety and depression in response group. The scores of HAMA,HAMD and digestive symptoms in response group after conventional therapy were significantly decreased(P < 0. 05),while the scores of HAMA and HAMD in refractory group after therapy were significantly higher than those in response group before therapy(P < 0. 05). The scores of HAMA,HAMD and digestive symptoms in combined therapy group were significantly lower than those in conventional therapy group(P < 0. 05). Conclusions:Relationship is found between FGIDs and psychological factors, and antidepressants might contribute to the amelioration of symptoms of FGIDs.

ABSTRACT

Objective To investigate the expression of transforming growth factor beta (TGF-β) and Smad signaling in ankylosing spondylitis (AS) and to explore their roles in the pathogenesis. Methods Fiftythree patients with AS were included in the study. In these 53 cases, 30 patients were performed computed tomography-guided needle biopsy in sacroiliac joint. Serum TGF-β_1 was determined by enzyme-linked immunosorbent assay (ELISA). Immunohistological studies were performed with the streptavidin-peroxidase conjugated methods to assess the expression of TGF-β_1, p-smad3 and Smad7 in sacroiliac joint tissue sample.One-way ANOVA, two independent samples t test and kolmogoorov-Simimov test were used to do statistical analysis. Results In 53 cases patients with AS, 20 cases were with high level Erythro-cyte sedimentation rate(ESR) and C-reactive protein (CRP), while those of the other 33 cases were normal. Serum average TGF-β_1level [ (15.9±5.6) ng/ml ], in patients with high level ESR/CRP [(5.4±5.8) ng/ml ] was significantly increased as compared to the controls and patients with normal ESR/CRP [(4.1±3.6) ng/ml] (P<0.05). There was no expression of TGF-β_1 could be detected in the pannus and bone marrow in SI joints tissue of 30 cases with AS, while decreased level of smad7 expression was detected. In addition, p-smad3 expression was found in the nuclear. Conclusion TGF-β_1 signaling may play an important role in the inflammatory erosion and cartilage fibrosis of sacrojlitis in AS.

ABSTRACT

The X-ray radiograph, CT scan and MRI appearance of 5 patients with pathologically proven fibrous dysplasia in thoracic and lumbar spine vertebrae were retrospectively analyzed. Plain radiographs, CT scans and MR images showed the presentation of eccentric lesion with intact cortex bone and marginal sclerosis in vertebral bodies without involvement of vertebral appendix and ex- traosseous soft tissue. The lesion masses were round (one being oval-shaped) and radiolucent in plain radiographs and CT scans. Homogeneous long signal was observed on TI weighted image and strongly enhanced when gadolinium was administered. On T2 weighted MRI, short signal was found in the anterior part of the mass, long signal in the posterior part, and short and slight long signal in the middle part, without partitioning and laminating change. There was a good correlation between ra- diological features and surgical findings. These findings may be useful to diagnose fibrous dysplasia in spine.

ABSTRACT

[Objective]To analyse the imaging findings of thoracic and lumbar fibrous dysplasia and evaluate the imaging characteristics.[Method]Among 5 cases of monostotic fibrous dysplasia of thoracic and lumbar spines,4 were imaged with CT scaning,2 with MRI and 5 with X-ray.All materials were analysed.[Result]Each X-ray revealed a round or oval radiolucent lesion surrounded by a distinct and thick rim of sclerosis.CT showed a round or oval low-density lesion with a high-density borderline between the lesion and normal vertebral bone tissue.Bone cortex remained intact in all cases. The lesion showed homogeneous long T1 signal on T1WI and hypo-or isointensity signal on T2WI.There was a characteristic hypointensity loop surrounded the lesion on T1WI,T2WI and PdWI.The lesion was demonstrated obviously in the contrast enhancement of scanning.[Conclusion]Monostotic fibrous dysplasia of thoracic and lumbar spine shows distinctive imaging characteristics that are different from dysplasia of limb bone or skull. These characteristics are decisive in the diagnosis of the lesion.
